5 out of 5 stars So brilliant that I cried--twice!.......2003-04-25

This is without a doubt the most brilliant and poignant depiction of HPLC and CE of biomolecules in modern history. My extreme hyperactivity disorder usually means that I'm not able to sit still for such weighty topics--not without large amounts of abused Ritalin; but Cunico's work was so compelling and finely wrought that I found myself spellbound, even when drunk, even when driving in the backseat of a stolen car across the Mexican border at Tecate. I am not going to recommend drinking heavily and stealing cars. But I do recommend BASIC HPLC and CE OF BIOMOLECULES by Robert L. Cunico. By the way, I am extremely drunk right now.

5 out of 5 stars Basic HPLC and CE of Biomolecules.......2000-12-24

This is a book that should be on the desk of anyone who is involved in analytical development of biomolecules and biopharmaceuticals. The content is well organized and written in a format that is easy to grasp and understand. The content covers almost all the different modalities of HPLC analyses and applications and provides real life examples of the techniques. It includes innumerable suggestions on how best to develop a validatable HPLC method and analyze the data appropriately.

I would highly recommend this book to both new and old members of biotechnology, especially the developmental, analytical, and quality assurance groups where there is frequently a need to implement a solid reliable method for analyzing biomolecules in a more rational fashion.

                              Description:
The retention behaviour of compounds in a chromatographic system is believed to be multivariate by nature, i.e. many physico-chemical properties of an analyte can influence its retention. Principal component analysis (PCA) and partial least squares (PLS) can therefore be particularly useful tools for visualising, exploring and modelling the complex interactions between solutes and the mobile and stationary phase. PCA allows the relationships between compounds (the observations) and their retention parameters (the variables) to be visualised in usually just two or three dimensions. PLS can be used to model quantitative structure-retention relationships (QSRRs) and may lead to better understanding of retention and selectivity changes in chromatographic systems. The objective of the study was to investigate the chromatographic behaviour of basic compounds under optimised gradient conditions using octadecyl high performance liquid chromatography (HPLC) columns designed for high pH separations. Three pharmaceutical mixtures were analysed by linear gradient reverse phase HPLC (RP-HPLC) at high pH using ammonia as a pH modifier, and methanol and/or acetonitrile as the organic modifier. The separations were carried out on three octadecyl columns: Waters XTerra MS C18, Agilent Zorbax Extend C18 and Thermo Hypersil-Keystone BetaBasic-18. Multivariate PCA and PLS modelling were employed to explore and explain the differences in selectivity between the chromatographic systems studied when the basic compounds were analysed under the high pH conditions. The interactions between the analytes and the mobile-stationary phases were described by relating the compound molecular descriptors with the selectivity of each chromatographic system. The selectivity differences between the chromatographic systems were identified.

                                  Description:
Three n-octadecylphosphonic acid-modified magnesia-zirconia reversed stationary phases (C"1"8PZM) are prepared via the strong Lewis base interactions between organophosphonate and magnesia-zirconia composite. And two of them are end-capped by using trimethylchlorosilane as end-capping agent in different procedures. Stability studies at extreme high pH conditions (pH 9-12) show that both the non-endcapped and endcapped columns are quite stable at pH 12 mobile phase. The reversed-phase liquid chromatographic behavior of three C"1"8PZM stationary phases are comparatively investigated in detail using a variety of basic compounds as probes. The retention of basic compounds on the three phases is studied over a wide range of pHs. And the possible retention mechanisms of basic compounds on the three stationary phases are discussed. The results show that the basic solutes retain by a hydrophobic and cation-exchange interaction mixed mechanism on three stationary phases when they are operated in eluents at pH values near to the pK"a of the Bronsted conjugate acid form of the analyte, suggesting that inherent zirconol groups on ZM are not expected to interact with bases via cation-exchange interaction at lower pH. Nonetheless, the non-endcapped phase differs markedly from the edncapped ones in retention and selectivity of basic solutes using eluents at pH 4.1, implying a complex retention mechanism at this pH. The cation-exchange sites under such conditions are more likely due to the adsorbed Lewis base anionic buffer constituents (acetate) on accessible ZM surface sites than the chemisorbed phosphonate. Although the three phases exhibit very similar chromatographic behavior with eluents at pH 10.1, and show in general satisfactory separation of basic compounds and alkaloids studied, the performance for a specific analyte, however, differs largely from column to column.
